A DNA probe relies on molecular hybridization. When mixed with other DNAs, they hybridize with strands that have a complementary sequence of nucleotide bases. To make a probe for a particular agent, workers isolate and amplify an appropriate segment of the organism's DNA by cloning in bacterial cells. The cloned DNA is denatured to make a single-strand probe, which is labelled with a radioactive isotope. When the probe is applied to a culture of material taken from a patient, it hybridizes with complementary strands. The fact of hybridization, which indicates that the organism cultured from the patient is indeed the organism that supplied the probe, can be revealed by autoradiography or other techniques.
